The main clinical manifestations include shortening of the limbs, severe shortening of the pelvis and femurs, growth abnormalities and spinal deformities, as well as possible abnormalities in joint function. The disease may also be accompanied by scoliosis, chest abnormalities, and excess adipose tissue. Otospondylomegaepiphyseal dysplasia is caused by genetic mutations that affect the development of the musculoskeletal system and the metabolism of cartilage tissue.

<div id="ez-toc-container" of the disease and interesting historical facts

Otospondylomegaepiphyseal dysplasia was first described in the mid-20th century. The main works on the classification and understanding of this disease belonged to researchers studying the genetic aspects of growth and development. Scientists noted the similarity of clinical manifestations with other forms of dysplasia, which led to difficulties in diagnosis. An interesting fact is that many studies of early cases of otospondylomegaepiphyseal dysplasia revealed the presence of this pathology in members of the same families, which indicated the hereditary nature of the disease.

Epidemiology

The epidemiology of otospondylomegaepiphyseal dysplasia demonstrates its extremely rare occurrence. According to available data, the incidence is approximately 1 case per 1 million births. This makes the disease one of the rarest pathologies of the osteochondral system. Gender differences in incidence are not observed, but a more frequent manifestation of the disease is noted in certain ethnic groups, which may be associated with the peculiarities of genetic predisposition.

Genetic predisposition to this disease

Otospondylomegaepiphyseal dysplasia is associated with mutations in various genes that are responsible for the development of cartilage and bone tissue. The most studied genes include COL2A1, SLC26A2, and GDF6. These genes are involved in the biosynthesis of collagen and other components of the cartilage matrix. Mutations in these genes can lead to disruption of normal growth and formation of cartilage tissue, which in turn causes clinical manifestations of the disease. Transmission of the disease is carried out in an autosomal dominant manner, which means that hereditary predisposition can be passed from one parent to offspring.

Risk factors for the development of this disease

Although the main risk factor is genetic predisposition, there are other physical and chemical factors that may influence the development of otospondylomegaepiphyseal dysplasia.
