The main microscopic changes are associated with a lack of bone matrix and impaired mineralization, which in turn leads to increased bone fragility and susceptibility to fractures. The disease often manifests itself in childhood, but may also remain unrecognized until adulthood. Clinical manifestations vary from minor dysplasia to severe forms accompanied by multiple fractures and patient disability. Treatment of the disease often requires an interdisciplinary approach, as it is associated with several specific clinical and laboratory diagnostics, as well as the need for continuous monitoring of the patient's condition.

<div of the disease and interesting historical facts

Osteoglobinic dysplasia was first described in the scientific literature in the mid-20th century. Research conducted by various groups has established that the disease may be associated with genetic mutations in a number of key genes. Interestingly, over time, many unique cases have emerged in medicine that have attracted the attention of researchers. One such case was described in 1973 papers, which presented a case of a family in which high rates of predisposition to osteoglobinic dysplasia were observed over several generations. This discovery gave rise to further research into the genetics and molecular biology of the disease. In recent years, the attention of researchers has been drawn to possible interactions between genetic and environmental factors influencing the development of osteoglobinic dysplasia.

Epidemiology

The epidemiology of osteoglobion dysplasia depends largely on ethnicity and geographic region. According to various data, the prevalence of this pathology varies from 1 in 100,000 to 1 in 250,000 newborns. At the same time, there is a significant genetic predisposition, and in some populations, cases of the disease may be observed significantly more often, which is explained by the high prevalence of mutations in certain families and populations. Studies show that men and women suffer from the disease with approximately the same frequency. The difficulty in establishing accurate statistics lies in the insufficient diagnosis of the disease, especially in childhood, which can lead to underestimated statistical data on the incidence.

Genetic predisposition to this disease

Osteoglobinic dysplasia is associated with mutations in a number of genes responsible for the processes of bone formation and mineralization. The main genes noted in this disease are COL1A1 and COL1A2, which encode collagen proteins. These proteins are the main components of the extracellular matrix of bones. Mutations in these genes lead to incorrect formation of collagen fibers and, consequently, to increased bone fragility. Rare mutations in other genes responsible for the regulation of bone metabolism, such as SP7 and RUNX2, have also been noted, indicating a multifactorial nature of the disease.

Risk factors for the development of this disease

Risk factors for osteoglobion dysplasia include both genetic and exogenous influences.
