The key symptom of the pathology is microphthalmia, a decrease in the size of the eyeball, combined with other ophthalmological disorders such as coloboma of the iris, cataracts, and glaucoma. In addition to ocular manifestations, the syndrome is accompanied by multiple systemic anomalies affecting the skeletal system, kidneys, cardiovascular system, and other organs. According to research, the disease demonstrates pronounced clinical polymorphism even among members of the same family.

<div id="ez-toc-container" class="ez-toc-v2_0_88 counter-flat of the disease and interesting historical facts

The first detailed description of the syndrome was presented by the German ophthalmologist Walter Lenz in 1955, after whom the syndrome was named. It is interesting to note that the disease was initially considered exclusively as an ophthalmological pathology, but subsequent observations revealed its multisystemic nature. In the 1980s, thanks to the development of genetic research methods, it was possible to establish a link between Lenz syndrome and mutations in certain genes. A significant contribution to the study of the disease was made by the work of Finnish scientists, who discovered a high frequency of the syndrome in some isolated populations of Northern Europe.

Epidemiology (statistics of disease occurrence)

According to current epidemiological data, the incidence of Lenz microphthalmia syndrome is approximately 1:50,000 newborns. However, this figure may vary depending on the region and ethnicity. Studies show an increased prevalence of the disease in some isolated populations, for example, in Finland, where the incidence reaches 1:40,000. “Sex distribution data show a predominance of the disorder in males,” the experts note in a paper published in the Journal of Medical Genetics (2019). Mortality at an early age is about 30% cases, mainly due to severe congenital malformations.

Genetic predisposition to the disease (involved genes and mutations)

Lenz syndrome has an X-linked recessive pattern of inheritance and is associated with mutations in the BCOR gene, located on the long arm of the X chromosome (Xp11.4).
