This type of cancer is characterized by an aggressive course, but its deviation from HER2-positive cancer gives a number of differences in epidemiology, pathogenesis and treatment methods. HER2-negative cancer is often associated with higher levels of estrogen and progesterone, but not all patients require aggressive therapy, which makes correct diagnosis and an individualized approach of the disease and interesting historical facts

Breast cancer has been the subject of intense medical research throughout the twentieth century. In the 1970s, scientists discovered that some tumors have specific receptors that can predict the response to hormonal therapy. In the 1980s, HER2 began to be studied as a marker in breast carcinoma. In 1990, the first drug targeting HER2 was approved, ushering in a new era of cancer therapy. However, until the 2000s, research on the HER2-negative form of the disease was not widely publicized, and there were relatively few studies on this subtype.

Epidemiology

HER2-negative breast cancer accounts for about 75% of all breast cancer cases. It is more common in women over 50 years of age and less common in younger women. Research shows that the risk of developing HER2-negative cancer is directly related to age, with the highest number of cases recorded in women aged 45 to 65 years. Given the global statistics, in recent years there has been an increase in the incidence among various populations, but these rates may vary in different countries.

Genetic predisposition to this disease

Genetic predisposition plays a key role in the pathogenesis of HER2-negative breast cancer. The main genes associated with the risk of the disease include BRCA1 and BRCA2, mutations in which increase the likelihood of breast cancer. These genes are involved in the process of DNA damage repair, and their functional impairment leads to the accumulation of mutations. Other important genes, such as TP53 and PTEN, can also significantly affect the development of the disease. Studies show that the pathogenetic mechanisms in the HER2-negative form have many directions, but, unlike HER2-positive cancer, their influence is often associated with hormonal activity.

Risk factors for the development of this disease

There are several key risk factors associated with the development of HER2-negative breast cancer:

Age: The risk of the disease increases with age, especially after 50 years.

Family history: Having a family history of breast cancer significantly increases your risk.

Hormonal factors: Long-term hormone replacement therapy and high estrogen levels may increase the likelihood of developing the disease.

Lifestyle: Being overweight, sedentary and drinking alcohol may also be risk factors.

Previous conditions: Having a history of other cancers or precancerous conditions increases your risk.

Diagnosis of this disease

Early detection of HER2-negative breast cancer is important for treatment success.
